Chondroitin 4-sulfotransferases, such as CHST11, catalyze the transfer of sulfate from 3-prime-phosphoadenosine 5-prime-phosphosulfate to position 4 of N-acetylgalactosamine residues in chondroitin .The deduced 352-amino acid protein has a calculated molecular mass of 41.5 kD. C4ST1 has an N-terminal transmembrane domain and 4 regions of homology with other sulfotransferases: a 5-prime-phosphosulfate-binding site, a 3-prime-phosphate-binding site, and 2 alpha-helical domains near the C terminus. Northern and dot blot analyses showed that C4ST1 was highly expressed in leukocytes and hematopoietic tissues.C4ST1 expressed by transfected Chinese hamster ovary cells increased incorporation of radiolabeled sulfate into chondroitin, chondroitin 4-O-sulfate, chondroitin 6-O-sulfate, and desulfated dermatan sulfate.
