CCDC106 repressed p53-mediated activation of a reporter gene in a dose-dependent manner. Western blot analysis revealed that CCDC106 repressed p53 activity by inducing p53 degradation, which was reversed by an inhibitor of the ubiquitin-proteasome degradation pathway.All 3 variants encode the same 280-amino acid protein, which has a calculated molecular mass of 32 kD and contains a bipartite nuclear localization signal. Western blot analysis detected an approximately 32-kD protein in all 4 human cell lines examined. Fluorescence-tagged CCDC106 accumulated in the nucleus of transfected HeLa cells.CCDC106 gene contains 6 exons. Exon 2 contains 3 alternative splice acceptor sites, and it has a translation initiation codon near its 3-prime end.
