The deduced 469-amino acid protein contains 2 predicted nuclear localization signals and a number of protein kinase superfamily subdomains including a conserved ATP binding region, a serine/threonine kinase signature motif, several conserved phosphorylation sites, and the PFTAIRE motif that is similar to the conserved Cdk motif PSTAIRE.
PFTK1 shares 95% amino acid sequence identity with the mouse ortholog, and 61% and 53% identity with the catalytic regions of PCTAIRE1 (PCTK1) and CDC2, respectively. Northern blot analysis detected a 6-kb PFTK1 transcript at highest levels in brain, pancreas, kidney, heart, testis, and ovary, and at lower levels in placenta, lung, liver, skeletal muscle, prostate, small intestine, colon, and peripheral blood leukocytes.
