EH Domain Binding Protein 1 (EHBP1)
The deduced 1,196-amino acid protein has a calculated molecular mass of 136 kD. It contains an N-terminal region with 5 closely spaced EH domain-binding NPF motifs, a central calponin homology (CH) domain, and a C-terminal CAAX box. EHBP1 associated mostly with the cytosol and membrane fractions of transfected COS-1 cells, and it colocalized with EH domain-containing protein-2 (EHD2) in mouse adipocytes.
NPF motifs of EHBP1 bound the EH domain of EHD2. Overexpression of the EH domain of EHD2 or the NPF motifs of EHBP1 disrupted cortical actin structures, and depletion of Ehd2 or Ehbp1 in mouse adipocytes by small interfering RNA inhibited endocytosis.
