Heparan Sulfate-3-O-Sulfotransferase 5 (HS3ST5)

HS3ST5 belongs to a group of heparan sulfate 3-O-sulfotransferases that transfer sulfate from 3-prime-phosphoadenosine 5-prime phosphosulfate (PAPS) to heparan sulfate and heparin. HS3ST5 expressed by transfected Chinese hamster ovary (CHO) cells displayed sulfotransferase activity toward heparan sulfate, generating at least three 3-O-sulfated disaccharides. HS3ST5 expressed by transfected Chinese hamster ovary (CHO) cells displayed sulfotransferase activity toward heparan sulfate, generating at least three 3-O-sulfated disaccharides. Transfection of HS3ST5 also rendered CHO cells susceptible to infection by herpes simplex virus-1, and HS3ST5-modified heparan sulfate bound the viral envelope protein, glycoprotein D. HS3ST5-modified heparan sulfate bound antithrombin, suggesting that HS3ST5 produces anticoagulant heparan sulfate.
