Histidine Triad Nucleotide Binding Protein 1 (HINT1)
PRKCNH1 is a member of a family of small proteins found in several species that inhibit PKC activity in the presence of the 14-3-3 proteins.the PKCI1 protein interacts with the regulatory domain of PKC-beta in the yeast 2-hybrid system. In the same system, PKCI1 also interacts with the ataxia-telangiectasia group D protein, which is mutant in ataxia-telangiectasia . PKCI1 was a potent inhibitor of PKC.
The authors commented that the protein is a member of the HIT family of proteins; the term HIT refers to the histidine triad near the C terminus in this family (i.e., his110, his112, and his114 in the human PKCI1 sequence). They noted that FHIT, a putative tumor suppressor gene, is a PKCI1 homolog.
