MED28 has an N-terminal proline-rich region, an N-glycosylation site, 4 phosphorylation sites, and 2 N-myristoylation sites. Northern blot analysis detected transcripts of 2.4 and 1.2 kb in HUVECs. Expression was also high in liver, placenta, and testis. In situ hybridization detected EG1 staining in arteries, veins, and capillaries, as well as in spleen endotheliocytes, placental Hoffbauer cells, hemangioma blood vessels, and epithelial cells of many organs. No EG1 was detected in lymphoid tissues, muscle, or fat. In a mouse catecholaminergic neuron cell line, magicin associated with the detergent-insoluble cytoskeletal fraction and in submembranous regions of cells, particularly the motile regions, such as leading or ruffling edges, where it colocalized with actin and merlin. It also localized to the actin cytoskeleton within neurites.
