RASSF6 interacted with RasGTP and bound K-Ras (KRAS) in a GTP-dependent manner via its effector domain. Coimmunoprecipitation studies demonstrated RASSF6 binding to proapoptotic protein MOAP1. Cotransfection of RASSF6 and KRAS showed a synergistic activation of cell death, and RASSF6 induced apoptotic cell death. SiRNA knockdown of RASSF6 expression in H1792 human lung tumor cells caused increased tumorigenicity as determined by increased growth in soft agar. Cancer profiling array showed that RASSF6 expression was downregulated in 30 to 60% of multiple tumor-derived tissues compared to normal tissues. RASSF6 transfection suppressed the levels of an NFKB (NFKB1) reporter, suggesting that RASSF6 may act via inhibition of the NFKB pathway.
