BCA2 encodes an ORF of 304 aa and contains various distinct domains: a RING-H2 finger, a zinc finger, an AKT phosphorylation site and several SH3 domain binding motifs.Ubiquitination of wild-type, RING-H2 and AKT site mutants of BCA2 was tested in HEK293T cells transfected with Flag-tagged expression vectors. After immunoprecipitation with Flag-antibody the wt and mt BCA2 proteins were incubated with E1 and E2 enzymes and Ub, and analyzed by Western blotting.BCA2 maps to chromosome 1q21, which has been implicated in breast and other cancers.
BCA2 is a novel RING-type E3 ligase that is important for the homeostasis of proteins crucial to breast carcinogenesis and that BCA2 and partner proteins may provide novel targets to develop effective treatments for breast cancer.
