The deduced 988-amino acid protein contains an N-terminal signal sequence, followed by 10 EGF-like repeats and a C-terminal CUB domain. It has 6 putative N-glycosylation sites in its C-terminal half. The mature 966-amino acid protein has a calculated molecular mass of 106 kD. RT-PCR showed that SCUBE1 expression was restricted to highly vascularized tissues, such as liver, lung, and kidney. In situ hybridization revealed SCUBE1 in microvascular endothelial cells of human umbilical cord artery and vein and of monkey brain, lung, and kidney. Western blot analysis of transfected human embryonic kidney cells detected SCUBE1 in the culture medium and associated with the cell membrane. Differential extraction revealed that SCUBE1 is a peripheral membrane protein.
