Hypotaurine uptake into rat placental brush border membrane vesicles and HEK293 cells expressing mouse was measured using LC-MS/MS. GABA at 100 μM significantly inhibit hypotaurine uptake, although taurine (100 μM) and betaine (2 mM) didn't inhibit it. These characteristics were comparable with characteristics of Slc6a13/GAT-2 and Slc6a11/GAT-3, rather than Slc6a1/GAT-1, Slc6a12 /BGT-1 and Slc6a6/TauT. Slc6a13 expression in rat placenta, although Slc6a11 was not detected. Hypotaurine uptake into HEK293 cells expressing mouse Slc6a13 was significantly higher than that into mock cells, and the inhibitory characteristics of hypotaurine uptake via mouse Slc6a13 was similar to into BBMVs. Ezrin expression in placenta is involved in the maintenance of hypotaurine concentration in fetus during the fetal development and Slc6a13 plays the role.
