Oh et al. (1997) cloned mouse Spin from a 2-cell stage embryo library and showed that it is homologous to the multicopy Ssty gene. In situ hybridization and RT-PCR assays detected murine Spin transcript in unfertilized eggs and 2-cell stage embryos, but not in 8-cell stage embryos.
Using differential display analysis of normal and cancerous ovarian tissues, followed by database analysis and PCR of total RNA from an ovarian cell line, Gao et al. (2005) cloned human SPIN. The 237-amino acid protein has a predicted molecular mass of 27 kD and shares 98% homology with mouse Spin. The C-terminal portion of SPIN was required for nuclear localization in transfected COS-7 cells.
