SPRED2 is a member of the Sprouty/SPRED family of proteins that regulate growth factor-induced activation of the MAP kinase cascade.
deduced 411-amino acid protein contains an N-terminal Enabled (ENA)/VASP homology-1 (EVH1) domain, a central KIT-binding domain (KBD), and a C-terminal SPRY domain. Endogenous Spred2 localized to the membrane in rat PC12 cells, and mutation analysis determined that the C-terminal SPRY domain was required for plasma membrane localization.Inhibition appeared to occur through the formation of a Spred-Ras complex that inhibited activation of MAP kinase by suppressing phosphorylation and activation of Raf.
