SUV420H2 and the related enzyme SUV420H1 function as histone methyltransferases that specifically trimethylate nucleosomal histone H4 on lysine-20 (K20). Accumulation of trimethylated histone H3K9 was unaffected. The deduced 468-amino acid protein contains a central SET domain and 3 additional regions that are conserved in Suv420h orthologs from other species. Fluorescence-tagged Suv420h2 localized to pericentric heterochromatin in transfected mouse embryonic fibroblasts (MEFs).the carcinogenic potential of human breast cancer cell lines correlated with loss of DNA methylation, which was accompanied by increased expression of maintenance DNA methyltransferase DNMT1 and the methyl-binding proteins MECP2 and MBD2, decreased trimethylation of H4K20, and hyperacetylation of histone H4.
