Tyrosylprotein Sulfotransferase 1 (TPST1)
TPST1 consists of 5 exons (with exon 5 being noncoding), varying in size from 47 to 926 bp, and with introns varying in length from 3.0 to 75.8 kb. By mutation analysis, the authors excluded TPST1 as the causative gene for Shwachman-Diamond syndrome, which maps to the 7q11 region.
The human cDNA encoded a fully functional N-glycosylated enzyme with an apparent molecular mass of approximately 54 kD when expressed in mammalian cells. This enzyme defined a new class of Golgi sulfotransferases that catalyze tyrosine O-sulfation of many protein substrates involved in diverse physiologic functions including inflammation and hemostasis.
