Northern blot analysis detected 2 USP25 transcripts of 4.6 to 6.5 kb at low to moderate levels in all tissues tested except testis, where only the shorter transcript was detected. In situ hybridization analysis of fetal mouse brain showed a clear correlation of expression with proliferative neuroepithelial cells and postmitotic neurons. High expression was also detected in adult mouse testis.
A transcript containing exon 10B in addition to exon 10A was detected predominantly in testis, with lower levels in small intestine, spleen, and peripheral leukocytes. Exon 10B introduces an internal stop codon, resulting in a protein truncated at 361 amino acids. Exon 19B adds 38 amino acids to the protein.
