The tag antibody can be highly specifically combined with the corresponding tag fused polypeptide or protein, which is used for isolation, purification and analysis of the target protein. Let’s make a brief introduction of serial protein tag products.
HA tag protein. It is derived from the antigen determinant of influenza virus’ red blood cell lectin surface, taging sequence YPYDVPDYA.And it has 9 amino acids,with small influence on the spatial structure of the exogenous target proteins, both easy in constructing a tag protein fusing to the N or C terminal and being detected by AntiHA antibody and ELISA .
MYC tag protein. The MYC tag protein is a small label with 11 amino acids(Glu-Gln-Lys-Leu-Ile- Ser-Glu-Glu-Asp-Leu), as the antigenic epitope expressed in different protein frameworks to identify their corresponding antibodies. Myc tag has been successfully applied in Western-blot hybridization, immunoprecipitation and flow cytometry to detect the expression of recombinant proteins in target cells.
FLAG. Flag tag protein is a hydrophilic polypeptide, consisting of 8 amino acids(DYKDDDDK),meanwhile the Kozak sequence constructed by the vector stimulate more efficient expression of the fusion protein with FLAG in eukaryotic expression system.
GST. Glutathione S-transferase plays an important role in the detoxification process, and its natural size is 26KD. As a highly soluble protein, GST can increase the solubility of exogenous proteins. In addition, the GST fusion expression system is widely used in the expression of various fusion proteins, which can increase the expression of the protein. The GST tag protein can be purified directly from the bacterial lysis solution using affinity resin that contains a reduced glutathione agarose gel (Glutathione sepharose) . Moreover, the GST tag can be eluted under mild/non denaturing conditions to keep the protein's antigenicity and biological activity. However, GST may loss the ability to bind the glutathione resin under denaturing conditions, and it’s better not to add strong denaturants, such as guanidine hydrochloride or urea, into the purification buffer.H6. His-Tag (HHHHHH),with 6 histidine tag, can be inserted in the C or N terminal of the target protein. It will not affect the target protein’s function because small molecular weight. And it can be purified under the condition of the presence of denatured and non ionic surfactants, As it has no steric hindrance, commonly used in interaction study among protein-protein and protein -DNA.
