Modeling Method
Preuer (Preyer) reflected normal guinea pigs, weight of 300 ~ 420g, the first otoscopy, excluding the external and middle ear disease, were tested 0.5,1.0,2.4 kHz 100 dB short pure tone ear reflex valve. Intraperitoneal injection of pentobarbital sodium was injected at 30mg / kg body weight. After anesthesia, the right external auditory canal was cleared and the external auditory canal was disinfected with 2% iodine tincture and 75% ethanol. 3X108 CFU / ml inactivated pneumonia Streptococcal suspension 0.1ml by tympanic membrane before and below into the animal tympanum, before injection culture on the agar medium to ensure that no live strain growth, to prevent acute infection. The model animals were examined by ear examination at 2 to 10 days after the suspension of the injected bacteria, and the color change of the tympanic membrane was observed. Followed by animal anesthesia after the death, take bilateral listening to the bubble, fixed with formaldehyde after stripping, paraffin embedded sections, conventional HE staining, light microscopy.
